Improve Purification Performance With High Capacities Using DurA Cycle A50
Addressing Modern mAb Purification Challenges
The latest protein A resin from Purolite, an Ecolab company, DurA Cycle A50 aims to maximize productivity and improve process economics.
DurA Cycle A50 improves purification performance with high capacities, which can help to enable a lower cost per gram of commercial drug product. Building on the innovation of Praesto™ Jetted A50 HipH, DurA Cycle A50 provides a wider elution pH window to address aggregate clearance.
Part of Purolite Affinity Resin Toolbox
DurA Cycle A50 is an agarose-based protein A resin designed to purify mAbs such as bispecifics and recombinant antibody fragments. It builds upon the innovation of Praesto™ Jetted A50, Praesto™ Jetted A50 HipH and Praesto™ 70 CH1.
The Purolite Difference
Developed in partnership with Repligen, an industry leader and partner in life sciences, DurA Cycle A50 provides high capacity and alkaline stability. Manufactured using Purolite’s patented Jetting technology, DurA Cycle A50 offers uniform beads to ensure quality and performance.
Features
- High Capacity 50 µm bead size
- Alkaline Stable
- Mild pH elution window
- High dynamic binding capacity of up to 80 g/L
Dynanic Binding Capacity
DurA Cycle A50 exhibits high dynamic binding capacities for conventional monoclonal antibodies, up to 80 g/l at a 10-minute residence time. Patented Jetting technology, coupled with the latest Protein A ligand technology, allows for high capacity even at low residence times. The resin is uniquely suited to address the demands of modern bioprocessing, from high-titer cell cultures to continuous chromatography formats.
Alkaline Stability
0.1 M Sodium hydroxide (NaOH) exhibits high efficiency in removing bound proteins, nucleic acids, and lipids from bioprocess resins, alleviating the risk of fouling on heavily burdened Protein A columns. Higher concentrations of sodium hydroxide may be used between campaigns and when more substantial cleaning is required.
DurA Cycle A50 is alkaline stable for over 60 hours of exposure to 0.5 M NaOH, and over 300 hours of exposure to 0.1 M NaOH.
Milder pH Elution Window
The ligand design allows for elution at a wider range of pH when compared to conventional Protein A chromatography. Allowing for a wider operational window to address aggregation concerns and increase purity clearance within the capture step.
