As upstream mAb productivity rises, downstream purification can become the limiting factor for throughput, cost, and facility capacity. An intensified workflow combines high-capacity Protein A capture, optimized AEX, HIC, and CEX polishing, inline buffer dilution, and integrated single-use chromatography to reduce operational burden while maintaining product quality. Modeled results indicate that this approach may reduce downstream footprint by approximately 30% and buffer volumes by roughly 79%, while capture-step yields can reach up to 95%. The use of concentrated buffers up to 10X can also reduce preparation, storage, and handling requirements.
