POROS Benzyl Ultra Viral Clearance And Impurity Removal For A Novel Antibody Format

Ambrx Inc, a firm that develops innovative Antibody-Drug Conjugates (ADCs) for cancer treatments, has been exploring the effectiveness of POROS™ Benzyl Ultra, a hydrophobic interaction chromatography resin, in reducing high molecular weight (HMW) species. These species are challenging to eliminate through standard bind and elute cation exchange chromatography due to their properties and high starting levels.
The application of POROS Benzyl Ultra demonstrated enhancements in scalability, performance, impurity reduction, and viral clearance abilities. The research was carried out using a monoclonal antibody with synthetic amino acids produced in recombinant CHO cell culture. The load material for POROS Benzyl Ultra had 7.5 – 10.2% overall HMW levels. The findings indicated successful recoveries, significant reduction of viruses, and impurity reductions.
