Scaling MSC manufacturing for clinical use depends on more than choosing the right supplement. The basal medium itself shapes expansion efficiency, and conventional options like alpha MEM leave significant yield on the table even when paired with human platelet lysate. This application note presents data showing that an MSC-specific, serum- and xeno-free basal medium supplemented with 5% hPL achieves at least 4-fold greater expansion than alpha MEM with equivalent supplementation, and outperforms multiple complete commercial MSC media across four passages.

Critically, these gains come without trade-offs: cells maintain characteristic surface marker profiles and retain trilineage differentiation capacity into chondrogenic, adipogenic, and osteogenic lineages.
