Enhancing mRNA-LNP Downstream Processing Via Single-Pass Tangential Flow Filtration
Single-pass tangential flow filtration (SPTFF) could provide a continuous alternative to conventional tangential flow filtration for downstream processing of mRNA lipid nanoparticles (LNPs). Comparative testing shows that SPTFF with 100 kDa regenerated cellulose cassettes can effectively concentrate mRNA-LNPs and remove ethanol while preserving key quality attributes, including recovery, particle size, and polydispersity. Notably, LNPs processed using SPTFF demonstrated 4.2-fold higher in vitro potency than those processed through conventional TFF. Although further investigation is needed, this result points to the possibility of achieving comparable biological activity with lower LNP doses, which could help limit toxicity risks associated with larger quantities.
