Enable Custom Chromatography Solutions For Downstream Processing

The study discusses the creation of custom ligand and resin platforms for manufacturing complex bio-therapeutics, including affinity resins. Custom ligand can be immobilized on a variety of backbones, including our high performing POROS™ beads. A case study the single step purification of prothrombin, showed the affinity resin had high selectivity for the most biologically active form of prothrombin, reducing host cell protein content in the eluate without compromising purity. The study also explored the development of a POROS prototype resin, investigating ligand immobilization, optimizing coupling conditions, and evaluating bead morphology impact on resin performance. The study found larger pore size resins had lower ionic and binding capacity due to lower surface area, while smaller pore sizes had high ionic but lower binding capacity. An increase in binding with ligand density was also observed.
