Developing A HIC Polishing Step For The Removal Of mAb Aggregates

In the downstream processes of mAb purification, mAb purity is high following the capture step. For this reason, it is crucial to remove impurities left after the capture step. To improve the purity even further, one or two polishing steps are commonly employed. The main goal of this study was to separate the target mAb from aggregates. Learn in-depth about the development of a flow-through (FT) polishing step that employs hydrophobic interaction chromatography (HIC) for the elimination of monoclonal antibody (mAb) aggregates. By using the Capto™ Phenyl High Sub resin, a reduction of aggregate content to below 1% was achieved with high monomer recovery, demonstrating that Capto™ HIC resins are an effective tool for aggregate removal for industrial mAb purification applications.
