Bispecific antibody manufacturing is genuinely hard, and multivalent molecules like those produced on Hinge Bio's GEM-DIMER™ platform make it harder still. The platform enables simultaneous targeting of multiple disease-associated antigens, with cooperative binding that drives higher biological activity than conventional monoclonal antibody approaches. That's a compelling scientific premise, but getting it to work at scale is another matter entirely.

The production challenges here were real: suboptimal cell growth, dissolved oxygen management inside the bioreactor, and a purification process complicated by the large size and multivalent structure of the molecules. None of these are trivial. Syngene's team worked through them by applying a Design of Experiments (DOE) methodology to identify the best combination of media, feeding strategy, feeding percentage, and temperature shift criteria, then refined aeration strategies to hit the necessary cell productivity targets.

The results are worth noting specifically: a three-fold increase in yield, 98.5% total purity, and overall recovery exceeding 50%, all completed within seven months. For a complex molecule program, that timeline is notably fast.
