What Is bYIok® Technology?
Developing IgG-like bispecific antibodies poses significant expression and product-quality challenges, particularly in achieving correct pairing of heavy and light chains. In this video, Lonza explores how its bYlok® platform is designed to address these challenges and enable more efficient bispecific development.
The precision-engineered bYlok® platform uses targeted modifications — relocating a disulfide bond to the VH/VL interface on one Fab arm — to promote correct heavy/light-chain pairing and prevent mispairing. Combined with established heavy-chain pairing technologies such as knob-in-hole, it enables greater than 95% full product assembly in Lonza's testing, in an IgG-like format that closely resembles a natural antibody while fitting into standard expression and downstream processes.
The video also shows how bYlok® fits into the development workflow — from upfront screening and molecule-specific format optimization through expression and purification — and outlines how customers can access the technology through licensing, in-house use, or early-development services.
Watch the full video to learn how bYlok® can improve bispecific antibody assembly, simplify development, and support manufacturability.
