5 Ways FATHOM EPR Complements NMR

Nuclear magnetic resonance spectroscopy provides valuable structural detail, but some biological questions require a broader view. Electron paramagnetic resonance spectroscopy can extend that view by measuring long-range distance distributions, distinguishing multiple conformational states, and detecting unpaired electrons in spin-labeled sites, metalloproteins, and radical intermediates. EPR can also generate structural information for intrinsically disordered regions, amyloid-like assemblies, and heterogeneous complexes that may produce broadened or missing NMR signals. These complementary insights support ensemble refinement, AlphaFold validation, PRE-constrained models, RDC analysis, and hybrid modeling with cryo-EM.
