As mRNA-based therapeutics continue to advance, analytical teams need reliable ways to characterize transcript size, purity, and consistency. Size exclusion chromatography offers a practical approach for separating in vitro transcribed mRNA across a broad size range, with retention time serving as a useful indicator of length for transcripts up to 4,000 nucleotides. Using a dSEC-7 column, the method separates mRNAs ranging from approximately 1,000 to 10,000 nucleotides and demonstrates a strong linear correlation between retention time and length in the 900–4,000 nucleotide range. The approach also provides visibility into sample purity, making it a valuable complement to capillary electrophoresis in mRNA analytical workflows.
