Unveiling Critical Translational Biomarkers Through Single-Cell Analysis

Donor heterogeneity significantly impacts the outcomes of adoptive cell therapies (ACTs). Traditional bulk measurements often fail to capture the nuances of cellular function that underlie diverse clinical responses. Single-cell functional analysis, as enabled by the Beacon platform, offers a more comprehensive approach to understanding these variations.
By combining cytotoxicity, cytokine secretion, and surface marker profiling, the Beacon platform identifies potential biomarkers correlated with patient outcomes. In this application note, we demonstrate how single-cell analysis can reveal predictive biomarkers for response, non-response, relapse, and adverse events like cytokine release syndrome or neurotoxicity.
A case study analyzing CAR-T products from four patients highlights the platform's ability to uncover distinct functional signatures associated with clinical outcomes. These findings underscore the potential of single-cell analysis to improve the precision of ACTs and pave the way for future multi-omic approaches that integrate genomic and transcriptomic data with functional performance.
