The Effect Of Mixing On Protein Using Mobius® Mixers

To address broad biomanufacturing mixing applications, we offer two distinct lines of single-use mixers with common attributes. This family of Mobius® single-use mixers uses magnetically-driven levitating impellers for improved mixing consistency and efficiency. The Mobius® MIX systems are designed for gentle mixing of solutions throughout the drug substance and drug product processes, whereas the Mobius® Power MIX systems are designed for vigorous mixing requirements in buffer and cell culture media preparation. Depending on individual manufacturing set-ups and organizations, mixing equipment may have to be used without distinction for all applications. Therefore, exploring suitability of both mixer types for model feeds which may be sensitive to mixing conditions is desirable.

Depending on users’ manufacturing set-ups and individual situations, Mobius® Power MIX and Mobius® MIX may have to be used without being dedicated to specific applications (for example using Mobius® Power MIX where gentle mixing may be sufficient).

To evaluate the effect of mixing in Mobius® MIX and Mobius® Power MIX on several model feed streams, a side-by-side comparison study was conducted. Various initial volumes and impeller speeds were tested to recommend best suited conditions. During each mix run, the protein quality was evaluated by periodically collecting samples to measure turbidity, perform SEC-HPLC (size exclusion chromatography) analysis, and evaluate filterability (Vmax™).
