SpinSep™

SpinSep™ is a new and simple procedure for isolating highly purified cells by negative selection without the need for magnets, columns, or other specialized equipment. Like our popular RosetteSep products for enrichment of human cells directly from whole blood, SpinSep can be used to enrich murine cells, and also to enrich human cells from samples other than whole blood, including previously processed and/or frozen samples. The SpinSep principle is similar to that of RosetteSep™. Unwanted cells in a cell suspension are made denser than the desired cells, allowing their separation via a simple buoyant density centrifugation step. Additional SpinSep™ enrichment cocktails will become available regularly. SpinSep™ does not require prior erythrocyte depletion. A cocktail of cell specific monoclonal antibodies is added directly to a splenocyte or bone marrow cells suspension, targeting the vast majority of unwanted cells for removed. Specially designed dense microparticles are then added and selectively bind the unwanted antibody-labeled cells, causing the overall density of the labeled cells to increase. The cell suspension is then centrifuged over the specially formulated density medium and the dense particle-labeled unwanted cells pellet, leaving the buoyant desired cells at the medium:buffer interface, where they are easily collected. Once the enriched cells are removed from the interface, they are immediately ready for use.
