This application note presents a scalable method for enhancing protein production using the ExpiCHO™ expression system in Thomson Optimum Growth® Flasks. Traditionally, scaling ExpiCHO™ beyond 500mL presented challenges. This protocol details a successful scale-up to 2L, improving workflow efficiency and space economy.

The study utilizes ExpiCHO-S™ cells cultured in OptiPRO™ SFM and transfected with ExpiFectamine™ CHO reagent. Key optimizations include:

Adjusted shaking speeds: Maintaining lower speeds post-transfection to enhance cell viability.

Controlled temperature shifts: Reducing temperature post-transfection to boost protein production.

Optimized humidity control: Minimizing evaporation and ensuring cell health.

Results demonstrate a significant increase in protein yield while maintaining comparable cell viability to standard methods. This scalable approach, coupled with the INFORS HT Multitron incubator shaker, offers a robust solution for maximizing protein production in the ExpiCHO™ system.
