Immobilized metal affinity chromatography (IMAC) is an effective chromatography tool for preparing recombinant proteins naturally rich in histidine/cysteine residues, as well as those with histidine residue cluster engineered to their N- or C-terminus via recombinant DNA technology. While a small quantity of protein prepared on a laboratory bench is often sufficient for structural and functional characterization studies, process-scale columns are needed in the manufacturing of recombinant proteins for therapeutic applications.
