Purification Process Development Of Plasmid DNA For Gene Therapy
By Ayushi Vasa, Kong Xiong, Hassan Sakhtah, Nida Zubairy, Tao Xiang, Chi-Ming Yu, Roy Lin, and Jaime De Souza

Plasmid DNA is a raw material for both mRNA and AAV processes, yet getting it into a pure, therapy-ready form remains difficult, beginning at lysis. This poster presents joint work from researchers at Boston Institute of Biotechnology and MilliporeSigma (Ayushi Vasa, Kong Xiong, Hassan Sakhtah, Nida Zubairy, Tao Xiang, Chi-Ming Yu, Roy Lin, and Jaime De Souza) on a reproducible, scalable approach to a 22 kbp plasmid, with the supercoiled isoform as the target. The data compare depth filter grades, calcium chloride dosing for RNA removal, 100 kDa and 300 kDa membranes in ultrafiltration/diafiltration, and membrane versus resin anion exchange capture, including purity figures for collected fractions. Study the pressure, flux, and gel data to see how each option performed.
