SYPRO Ruby is a one-step protein stain for the analysis of proteins in 1-D sodium dodecyl sulfate (SDS)–polyacrylamide or 2-D polyacrylamide gels with sensitivity equal to silver staining. The stain is fully compatible with subsequent microanalysis, such as mass spectrometry or Edman sequencing, and can be visualized with a simple UV transilluminator or a laser scanner. The gel is incubated in the staining solution for as little as 90 minutes. No stop solutions or destaining steps are required and gels can be left in the dye solution for indefinite periods without overstaining, simplifying the simultaneous processing of multiple gels. This gel stain shows little protein-to-protein variability in staining and can detect heavily glycosylated proteins, not detectable by silver staining. It also does not detect extraneous nucleic acids in the sample.
