Bispecific antibodies (bsAbs) can become unstable under typical Protein-A elution conditions (pH 3—3.5), causing aggregation and fragmentation. Novel Purolite Praesto Jetted A50 HipH resin is designed to elute antibodies in a higher pH range (up to pH 5). The unique properties of this resin allow for efficient elution at milder pH levels to conventional protein A chromatography. Two bsAb molecules were evaluated by Praesto Jetted A50 HipH and a Standard Protein A resin to maximize monomer purity. Praesto Jetted A50 HipH resin demonstrated increased separation and monomeric recovery when compared to the standard resin.
