NGS Identification Of Internal Tandem Duplications In FLT3 With Archer™Assays

This content is brought to you by Integrated DNA Technologies, a Danaher Operating Company.
Detecting FLT3 internal tandem duplications (ITDs) remains one of the more complex challenges in acute myeloid leukemia research, largely due to their variable length and repetitive structure. Leveraging Anchored Multiplex PCR (AMP) chemistry, Archer VARIANTPlex assays enable open-ended amplification and deep, bidirectional coverage of ITD regions without prior knowledge of insertion size or location. Combined with the Archer Analysis pipeline, this approach supports de novo assembly of ITD-containing sequences and more accurate allele frequency determination, even in low-input or degraded samples. Beyond ITD detection, the platform enables simultaneous profiling of multiple clinically relevant mutations across genes like NPM1, CEBPA, and TP53. For researchers seeking comprehensive genomic insight from a single workflow, this approach offers a scalable and sensitive path forward.
