StemSep™ is a negative selection system that immunomagnetically labels unwanted cells and binds them to a magnetic column. The desired cells, which are collected in the column flowthrough, have not had antibody bound to their surface and are suitable for further functional studies. The production of rare cells with negative selection involves the labeling and removal of the vast majority of cells in a start cell suspension. This labeling process is achieved by using unmodified colloidal magnetic dextran iron and non-covalent bi-specific antibody cross-linking reagents called tetrameric antibody complexes. The small size of the particles facilitates the delivery of the magnetic label to the cells, and the use of bispecific tetrameric antibody complexes ensure reproducibility and ease of scale-up.
