Accurately quantifying GLP‑1 receptor agonists at sub‑ng/mL levels remains a persistent challenge in bioanalysis, driven by low dose regimens, long half‑lives, and peptide interactions with metal surfaces. This application-focused study explores how moving from conventional 2.1 mm analytical columns to 1.0 mm microflow chromatography can meaningfully improve assay performance without adding operational complexity. Using a UHPLC‑MS/MS workflow, four widely used GLP‑1 therapies were evaluated to assess sensitivity, chromatographic integrity, and solvent usage. The results show that microflow columns can deliver a two‑fold increase in peak response while maintaining comparable separation quality and run times. Just as notably, solvent consumption was reduced by 75%, translating into tangible cost and sustainability benefits.
