LOTTE Biologics Tool-Box Approach On Bispecific Antibody Purification And ADC Conjugation Modalities
By Saumyadip Nemu, Sangeetha Subramanian, Kevin Namitz et al.

Bispecific formats and dual payloads raise the bar for conjugation control and analytical depth. Authored by Saumyadip Nemu, Sangeetha Subramanian, and colleagues at LOTTE Biologics, this poster presents a modular tool-box approach spanning antibody purification, linker-payload chemistry, and characterization. Two case studies put it to work. The first applies partial reduction and cysteine conjugation of a bispecific antibody with deruxtecan at a target DAR of 6, with SPR binding kinetics and conjugation-site confirmation by peptide mapping. The second pairs dual-linker conjugation with a click reaction to carry two payloads toward DAR 8, supported by subunit RPLC/LCMS, CE-SDS, iCIEF, and SEC data.
Examine the analytical results to see how each conjugate performed across the orthogonal methods.
