LC/MS Characterization Of GLP-1 Receptor Agonist Oligomers And Other Elusive Impurities

GLP-1 receptor agonist impurities can arise during synthesis, purification, and storage, and some, including oligomers and isomers, push LC/MS workflows to their limits. Rachel Franklin of Agilent Technologies presents a two-stage approach built on complementary instruments: fast screening to flag impurities, followed by targeted MS/MS for detailed sequence information. Using liraglutide samples that were force-oxidized or stored for 15 months, the work shows how fresh and aged material behave differently, how a high m/z filter combined with all-ions CID separates oligomer behavior from overlapping signals, and how guided de novo sequencing localizes an abundant impurity that did not match the database.
