Precise identification of conjugation sites is critical to understanding and optimizing antibody–drug conjugates. A streamlined workflow integrates automated protein sample preparation with high-resolution LC/Q-TOF mass spectrometry to map lysine-linked modifications with confidence. In a T-DM1 case study, 26 conjugation sites are identified, supported by strong mass accuracy and consistent reproducibility across runs.
