High-salt mobile phases used in hydrophobic interaction chromatography (HIC) can put significant strain on conventional LC systems, introducing risk of corrosion and compromised performance. A bioinert approach with a completely iron-free flow path enables reliable drug-to-antibody ratio (DAR) analysis under these demanding conditions, supporting accurate characterization of antibody-drug conjugates. Using brentuximab vedotin as a model, precise DAR determination (3.7) was achieved alongside strong reproducibility, reinforcing confidence in data integrity. Advanced software capabilities further improve analysis by correcting baseline drift associated with lower-purity reagents, simplifying integration and interpretation.
