Generation Of Stable CHO Cell Lines For Protein Production

Obtain exceptional yields of recombinant protein using the CHOgro® High Yield Expression System. Establish stable cell lines through selection and expansion of single cells or a pool of cells that continually produce high levels of recombinant protein. By following the five steps of a specialized protocol, stable cell lines can be achieved in CHO cells using antibiotic selection. Not all serum-free complete media formulations can support transient transfection. Thus, CHOgro® Expression Medium is recommended for use with this protocol. The steps include:

1. Generate a kill curve to determine the optimal selection antibiotic concentration
2. Transfect cells with desired plasmid construct(s)
3. Select and expand stable polyclonal population (mini-pools) and assess GOI expression
4. Select single clones from subset of polyclonal population
5. Assess expression of GOI and cryobank high-expressing clones
