Efficient Purification Of Diverse Single Domain Antibodies Using HyperCel Mixed Mode Resins

Single-domain antibodies (sdAbs), also known as NANOBODY® molecules, offer unique advantages in research and medicine, but their purification can be challenging due to limited affinity for Protein A. This study demonstrates how HyperCel mixed-mode resins—MEP HyperCel and CMM HyperCel—deliver efficient capture and high recovery of sdAbs with neutral and acidic isoelectric points. MEP HyperCel achieves dynamic binding capacities far exceeding traditional affinity and cation exchange resins, while maintaining low host cell protein and DNA levels. For acidic sdAbs requiring mild elution conditions, CMM HyperCel provides near-neutral pH recovery close to 100%. Both resins enable rapid processing under low-salt conditions, reducing costs and preserving protein integrity. These findings highlight scalable, economical solutions for diverse protein purification needs.
