A weak clone selected during early screening can create downstream challenges, leading to longer development timelines, more difficult scale-up, and manufacturing risks at commercial scale. To address this, scientists at the Boston Institute of Biotechnology evaluated three approaches for generating high-producing minipools: standard transfection, effective transfection, and efficient screening. Results showed that two of the three strategies achieved approximately 1.5-fold higher titers while reducing minipool development timelines by 50% compared with the standard method. The team then advanced five monoclonal antibodies through limiting dilution cloning, HTRF-based screening, and 14-day fed-batch production studies to assess clone performance and productivity. Multiple single-cell clones across the programs achieved titers approaching 5 g/L, demonstrating the potential of optimized screening workflows to improve cell line development outcomes.

Review the poster for detailed methodology, comparative titer data, and complete 14-day production results that highlight how early clone selection can shape manufacturing success. Could your current screening strategy be leaving productivity gains on the table?
