Faster, more cost-efficient plant genotyping is becoming essential as agricultural testing demands increase. Direct amplification approaches are reshaping traditional qPCR workflows by eliminating nucleic acid extraction—one of the most time-consuming and resource-intensive steps. Using strawberry leaf punch samples as a model, this work demonstrates how extraction-free qPCR can deliver reliable detection of resistance markers while significantly reducing turnaround time and per-sample cost.
Results show clear clustering and strong amplification even without purification, with optimal performance achieved when minimal sample cleanup is applied. The approach also highlights the balance between workflow simplicity and data clarity when working with plant particulate.
