Determining Antibody-Drug Conjugates Positional Isomer Distribution With Nonreduced CE-SDS And HIC
Drug-to-antibody ratio is only part of what determines how an antibody-drug conjugate behaves. Where the payload sits matters too, yet the routine characterization methods each leave a gap. HIC resolves DAR0 through DAR8 species but offers no positional information. Nonreduced CE-SDS quantifies the fragments produced when payloads displace interchain disulfides, pointing to drug location but not DAR. This session shows how data from both, acquired on the Agilent ProteoAnalyzer system and the Agilent 1290 Infinity III Bio LC, can be combined through a system-of-equations approach to resolve individual positional isomers. For teams working with structurally complex conjugates, it shows how two familiar measurements can answer a question neither answers alone.
