Data-Driven CHO Bioprocess Optimization And Scale-Up With At-Line Intelligence Of Key Media Components

Given the unique nutritional requirements of each CHO cell line, traditional fed-batch processes often face challenges related to overfeeding, which can lead to the accumulation of toxic metabolites such as lactate and ammonium, negatively impacting product quality and increasing costs.

This study aimed to reduce these issues by employing the REBEL Amino Acid Analyzer for at-line, rapid analysis of amino acids to fine-tune feeding strategies. This approach enabled the identification of optimal nutrient levels, improving cell viability, reducing toxic metabolite accumulation, and ultimately achieving a 30% increase in titer under optimized conditions.

The scale-up from microbioreactors to 10L bioreactors was successful, demonstrating the effectiveness of REBEL-guided feed strategies in maintaining consistent nutrient profiles and product quality. This data-driven approach not only optimized process performance but also reduced the cost of goods by minimizing the use of costly feed supplements.
