Comprehensive Workflow Of Intact And Subunit Mass Analysis For Monoclonal Antibodies (mAb)

This application note presents the straightforward analysis of the integrity of a mAB through intact and different subunit approaches, leveraging liquid chromatography coupled to high-resolution quadrupole time-of-flight mass spectrometry. Alongside mass confirmation, different post-translational modifications (PTMs) such as glycosylation and terminal modification were characterized. The heterogeneity and complexity of protein therapeutics have driven the adoption of mass spectrometry as one of the leading technologies for their characterization. 

Here, a comprehensive, yet straightforward, workflow of intact and subunit analysis for NISTmAb is reported using the SCIEX ZenoTOF 7600 system combined with BPV Flex software 2.1.
