Chemiluminescent Substrate

The DuoLuX Chemiluminescent Substrate is a formula based on acridan chemistry which allows one substrate to be used for either alkaline phosphatase or peroxidase detection. This substrate can be used for western, Southern, northern, or dot blots, or for ELISA applications with either enzyme as the detection system. The substrate using either enzyme has very high sensitivity and prolonged light emission characteristics. For developing Southern, northern, or dot blots with alkaline phosphatase conjugates, optimal film exposure times range from 30 seconds to 10 minutes with sensitivity less than 1 pg of nucleic acid. Using peroxidase as the detecting enzyme, exposure times are somewhat shorter with sensitivities slightly less than those for alkaline phosphatase. Both nitrocellulose or nylon membranes can be used for transfer, but nitrocellulose is the preferred membrane.
