BIOXYTECH GSH-420 Quantitative, Colorimetric Determination of Total Glutathione
Reduced glutathione (GSH) is an ubiquitous tripepticle (y-glutamylcysteinylglycine) which functions as a coenzyme in amino acid transport in detoxification of xenobiotics and carcinogens, In synthesis of DNA precursors and as an antioxidant1 Most GSH In whole blood Is found in the erythrocyte2.
Reagents
The reagent set is sufficient for 100 determinations and consists of the following Components:
- 1)Chromogen: 3 mM 1-Methyl-4-chloro-7-trifluoromethylquinolinium methylsulfate In 50 mM HCI. 20 mL.
2)Color Developer: 2 N Na0H in water, 20 mL.
3) Reducing Agent: 1,25 mM Tris(2-carboxyethyl)phosphine (TCEP) In 25 mM HCI, 20 ml
4)Precipitation Reagent: 7.5% (v/v) Trichloroacetic acid (TCA) in water, 2 x 25 ml
5) Buffer: 600 mM Potassium phosphate, 600 pM DiethyienetriamInepentascetic acid (DTPA), 0.09% Lubrol, pH 7.8, 20 mL
6) Calibrators: Low. 75 µM Glutathione disulfide (150 pM GSHI equivalents) in 126 mM potassium phosphate, pH 7.5, 3 ml High, 175 µM Glutathione disulfide (:)SO pM GSH equivalents) in 126 mM potassium phosphate, pH 7. 5, 3 mL.
