Accurate mAb Sizing Using The NIST mAb As A Ladder For The Agilent ProteoAnalyzer System
By Kyle Luttgeharm and Whitney Pike

Accurate sizing of intact monoclonal antibodies remains a persistent challenge due to their complex higher-order structures, often limiting confidence in quality control analyses. Integrating the NIST monoclonal antibody reference material (RM 8671) as a ladder within the Agilent ProteoAnalyzer workflow offers a practical path to improved measurement reliability. By anchoring sizing analysis to a well-characterized standard, variability is reduced, and confidence in molecular weight determination increases even for nonreduced mAbs. Results from multiple representative samples show meaningful gains in both accuracy and precision, supporting more consistent characterization across workflows.
For teams working to strengthen analytical rigor without adding unnecessary complexity, examine how this approach presents a straightforward enhancement to existing methods.
