Loading a column to 10% breakthrough is standard practice, yet it leaves a considerable share of binding capacity unused, which matters most for costly resins like Protein A. Periodic countercurrent chromatography addresses that waste, but it usually calls for specialized equipment. This poster from Boston Institute of Biotechnology, authored by Kady Oakley, Ph.D., and colleagues, shows how a traditional single column mAb purification was converted to a multi-column process on conventional instruments. It follows the method from a small-scale binding capacity test through the roles of upstream, midstream, and downstream columns, then compares yields from two runs and the adjustment that changed the second.

Explore the data and column setup to see how the conversion works.
