Touch-Free T Cell Expansion With A Fill-And-Forget Workflow

This experiment explores the use of G-Rex® bioreactors for large-scale, low-cost immune cell therapy manufacturing. The bioreactors are designed for a "fill-and-forget" workflow, eliminating manual interventions, reducing contamination risk, and streamlining manufacturing. The study compares this workflow with a traditional mid-point cytokine spike workflow, finding equivalent or improved T cell expansion, viability, and phenotypes, as well as reduced reagent and overhead costs. The G-Rex® bioreactors enable high-throughput manufacturing, further cutting costs and increasing flexibility. An experiment using isolated T cells was conducted, with the cells thawed, plated, and activated in a G-Rex® 6M culture vessel. Results showed that robust T cell expansion can be achieved with this workflow. The study also examined cytokine concentration in cell cultures over 9 days, finding that starting concentrations were not fully consumed during the culture period.
