Optimizing And Monitoring AAV Capsid Purification Using Mass Photometry
The SamuxMP mass photometer is a desirable system for optimizing and monitoring AAV (adeno-associated virus) downstream purification processes with technology that allows for accurate quantification of AAV loading, regardless of particle size or shape. It's ability to quickly measure the empty/full capsid ratio of AAV and detect impurities provides crucial information for process development. In this study, the SamuxMP was used to analyze AAV samples at various stages of purification, including after harvest and clarification, to confirm column binding, and to monitor elution from the chromatography column. The results showed the removal of impurities during clarification, the absence of particles in the AAV capsid mass range in the flowthrough, and variability in the percentage of full capsids during elution.
The SamuxMP offers advantages such as requiring minimal sample, quick measurement time, and low operational costs. Overall, the SamuxMP is an ideal tool for optimizing AAV purification processes.
