The effects on both IkBa phosphorylation and E-selectin were rapid, irreversible

BAY 11-7082 and BAY 11-7085 are two inhibitors of cytokine-inducible IKBa phosphorylation. Both drugs inhibit TNFa-induced phosphorylation and degradation of inducible IKBa in intact HUVECs without affecting constitutive phosphorylation.

The inhibition of IKBa phosphorylation and degradation results in the inactivation of NFKB. Both drugs inhibit the NFKB-dependent expression of endothelial-leukocyte adhesion molecules E-selectin, VCAM-1 and ICAM-1.

The effects on both IKBa phosphorylation and E-selectin were rapid, irreversible and occurred at an IC50 of approximately 10µM.
