A Thermo-Fast® 96 plate with carboxy (-COOH) groups allows covalent attachment of aminated oligonucleotides using a simple water soluble carbodiimide method. In a standard 50µl reaction volume about 5pmole of COOH groups are available for coupling up to 50ng of oligonucleotide.
Once the primer or probe is immobilised it will withstand the harsh conditions of a complete amplification program, retaining some of the PCR product on the walls of the tube for detection.
Carboxy Thermo-Fast® can be used to immobilise oligo(dT) for micro-isolation of nucleic acids with poly-A tails or any aminated oligo for specific hybridisation in a thermal cycler.
