Bioconjugates are molecules containing a targeting agent chemically linked to a payload, with at least one component being a biomolecule. Antibody-drug conjugates (ADCs) stand out as the most advanced bioconjugate therapies, particularly for oncology indications. ADCs employ monoclonal antibodies (mAbs) to transport highly potent APIs (HPAPIs) to targeted cells. HPAPIs exhibit more selective cytotoxicity in conjugated form. As a result, non-target cells are spared from the many toxic effects, thus improving the safety profile.

The current ADC pipeline encompasses emerging subclasses of bioconjugates that differ in antibody format (e.g. fragments or bispecifics) and payloads (e.g. non-cytotoxic small molecules, oligonucleotides, and chelators). The strategic combination of these components optimizes therapeutic outcomes.

MilliporeSigma's ADC & Bioconjugation Center of Excellence in St. Louis (USA) has over 35 years of expertise in bioconjugation, API, excipients, and adjuvants manufacturing.
